In my 10 years in science, I think I've never been to any research conference, on any topic, without hearing my colleagues interject dire warnings into their presentations – and I've never attended a climate-focused conference. To identify general trends in the observed epistatic interactions, we utilized unsupervised learning to elucidate patterns among the relative PME values of variants across each genetic background. We first utilized uniform manifold approximation projection (UMAP) (20) to identify mutations that have similar expression profiles across these conditions. A projection of the variants onto a resulting two-dimensional coordinate reveals that most variants fall into one of approximately three groups (Fig. 6A). Using a density-based hierarchical clustering analysis (HDBSCAN), we unambiguously assigned 182 variants into three distinct clusters based on their expression profiles alone (Fig. 6A, Table S4) (5). An analysis of the structural context of these mutants reveals that one of the largest clusters (Cluster 3, n = 76) primarily consists of mutations of soluble loop residues that have little impact on expression and exhibit minimal epistasis (Fig. 6 A-C). This analysis also identified a smaller cluster of loop mutations (Cluster 2, n = 30) that moderately decrease expression and exhibit a greater propensity to form epistatic interactions with V276T and/ or W107A relative to the neutral loop mutations in Cluster 3 (Fig. 6 A-C). Nevertheless, most of the mutations that exhibit strong positive epistatic interactions with W107A fell into Cluster 1 (n = 76), which primarily consists of mutations within TMDs that significantly decrease expression in all three genetic backgrounds (Fig. 6 A-C). A comparison across these clusters suggests positive epistatic interactions with W107A primarily stems from the fact that mutations that compromise expression can only cause a relatively modest reduction in the PME of W107A GnRHR (Fig. 6 B & C), which is already predominantly misfolded. In contrast, the moderately disruptive loop mutations in Cluster 2 tend to exhibit more pronounced negative epistatic interactions with V276T than do the highly disruptive TMD mutations in Cluster 1 (Fig. 6C). Generative AI can offer useful tools across the recruiting process, as long as organizations are careful to make sure bias hasn’t been baked into the technology they’re using. For instance, there are models that screen candidates for certain qualifications at the beginning of the hiring process. As well-intentioned as these models might be, they can discriminate against candidates from minoritized groups if the underlying data the models have been trained on isn’t representative enough. As concern about bias in AI gains wider attention, new platforms are being designed specifically to be more inclusive. With respect to the lateral part, it has been shown that it encodes negative reward prediction errors and is deeply involved in value-based decision-making such as active avoidance behaviors, but the neural pathway from the medial part to the interpeduncular nucleus has long been unknown, even though it is one of the most evolutionarily conserved pathways. Using adult zebrafish as material, we found that this homologous pathway is composed of two parallel pathways that antagonize each other to control emotional and decision-making behaviors, including social fighting behavior, with one pathway being used to predict interoceptive perception to realize a preferred posture for attacking an opponent or obtaining a reward and the other to attend to exteroceptive perception to evaluate whether the external world is safe or dangerous. In this talk, I will talk about our strategy to gain a complete picture of the neural circuit mechanisms of behavioral control by these pathways. Therefore, to support a claim of functional reorganisation, that is, novel local processing that supports novel behaviour, causal evidence is needed. In humans, a handful of TMS studies have reported that when processing in visual cortex of blind individuals was disrupted, perceptual abilities were also impaired. For example, TMS applied to the occipital pole doubled the errors made during Braille reading, relative to ‘air’ TMS (Cohen et al., 1997). Notably, the disruptive impact of the TMS mid-occipital stimulation was qualitatively greater than the impact of TMS stimulation over the sensorimotor (reading) hand territory itself. This raises a red flag, as any presumed reorganisation is not likely to have completely taken over the functional role of S1 in tactile processing: the contribution made by the visual cortex should not be greater than that of S1. TMS over V1 has since been shown to impair verbal semantic processing in blind individuals (Amedi et al., 2004), indicating that the disruptions in Braille reading may not be sensory at all. However, here, too, caution is needed. Considering that repetitive TMS will propagate its effects beyond the stimulation site (Bestmann et al., 2008), conclusive causal evidence in humans for the specific role of low-level visual cortex in supporting perceptual or cognitive abilities is still missing. Applying these in the context of the Human Pangenome Project yields the most complete picture of human sequence evolution available to date. And we are able to understand the last unexplored frontier of human genomes, the short arms of the acrocentric chromosomes. We show that in these regions, standard chromosome relationships break down, and the chromosomes form a community in which recombination occurs between heterologous chromosome pairs, a phenomenon not seen elsewhere in the human genome and never before characterized. The rise of knowledge and the flow of innovation have long been recognized as an important factor for economic development. How do increased spatial connections affect the generation and diffusion of ideas? Intuitively, a denser network should increase the number of novel ideas and augment their diffusion. I study knowledge production in Germany in the 19th century, relying on the universe of bibliographic records and novel railway statistics, among other original data, as well as cutting-edge machine learning and topology to measure ideas. I show that the railroad network increased the creation of new ideas, contributing to 11% of the increase in knowledge production. Scholars' mobility led to the formation of specialized clusters, and thus to cities' specialization. New ideas are formed by combining ideas coming from cities connected by the railroad. However, with a denser network, new ideas diffused less on average. This was a by-product of specialization: with the railroad, groups of scholars could focus on narrower topics and co-locate with similar professionals; they learnt more from similar groups, but became disconnected from dissimilar ones.
